Gonadotropin releasing hormone (GnRH) is a hypothalamic decapeptide that binds to GnRH receptors on pituitary gonadotrope cells to modulate the synthesis and secretion of the gonadotropins, luteinizing hormone (LH) and hormone (FSH)

Gonadotropin releasing hormone (GnRH) is a hypothalamic decapeptide that binds to GnRH receptors on pituitary gonadotrope cells to modulate the synthesis and secretion of the gonadotropins, luteinizing hormone (LH) and hormone (FSH). These gonadotropins in turn regulate gonadal steroidogenesis and gametogenesis. Chemical characterization and structure-activity analysis of GnRH variants containing systematic amino acid substitutions led to the discovery of GnRH superagonists and antagonists.

The antagonists abarelix and degarelix show low scores, based on a higher . Therapy with LH-RH analogues and antiandrogens achieved a complete clearance of lung involvement.

The antagonists abarelix and degarelix show low scores, based on a higher incidence of adverse events, a higher dosage frequency, more drug interactions and a more limited documentation compared with the agonists. The availability of a generic formulation of leuprorelin may lead to a reduction in cost.

This volume provides an overview of the mechanisms of action and clinical efficacy of gonadotropin (luteinizing) hormone releasing (LH-RH) agonists and antagonists in hormone-sensitive neoplastic diseasis. It contains a detailed description of structural and functional aspects and presents the latest clinical results in treating benign and malignant neoplasias with LH-RH agonists as well as a detailed review of present knowledge of LH-RH antagonists.
